Researchers find critical regulator to tightly control deadly pulmonary fibrosis

An international team of researchers led by Georgia State University scientists have found a key component in the pathological process of pulmonary fibrosis, a fatal disease for which there is currently no cure.

The scientists found that a key human gene, CLYD, serves as a crucial negative regulator in the development of the disease, halting its progression that leads to death. The research was published today in the journal Nature Communications.

"In some patients, CLYD does not function as it should or its protein level is lower than in normal individuals," said Jian-Dong Li, director of the GSU Center for Inflammation, Immunity and Infection (CIII) and Georgia Research Alliance (GRA) Eminent Scholar in Inflammation and Immunity.

"If this does happen, the human tissue repairing response can go out of control, leading to the development of fibrosis," added Li, senior author of the study and professor of biology at GSU.

According to the American Lung Association, about 140,000 Americans have been diagnosed with the disease. Patients' breathing symptoms worsen over time, and many patients live only three to five years after diagnosis.

There are currently no effective medicines available to health care professionals to cure pulmonary fibrosis. Professionals can treat the symptoms to reduce inflammation using steroids and immunosuppressants, but there are serious side effects over time, including immune system suppression, which makes patients even more susceptible to infections.

"The disease often develops after infection or injury. In the case of infections brought on by Streptococcus pneumoniae, a form of pneumonia, the body's immune system responds and tries to repair the damage, but in the case of fibrosis, this repairing process is overactive and causes scarring of the lungs," said Jae Hyang Lim, a GRA Distinguished Investigator and assistant professor at the CIII and Department of Biology.

The findings Li and his colleagues have presented have opened an exciting door to development of new therapeutics to fight the disease by aiming their sights on the CLYD gene.

"These results tell us that we now have a key regulator to target," said Binghe Wang, Director of the GSU Center for Diagnostics and Therapeutics and GRA Eminent Scholar in Drug Discovery and a co-author of the study.

"If we can target CLYD with a focus on regulating its protein level or activity, we may be able to better control fibrosis," said Wang, also chair of the Department of Chemistry at GSU. "There is a huge clinical potential in saving the lives of these patients."

The results are published in "CLYD Negatively Regulates Transforming Growth Factor-? Signaling via Deubiquitinating Akt," Nature Communications, DOI: 10.1038/ncomms1776. The journal is online at www.nature.com/ncomms/index.html.

Li said the support provided by the GRA is invaluable to the groundbreaking research performed by scientists on the project. Li was named as a GRA Eminent Scholar in 2010, and Lim came to Georgia State's CIII as a GRA Distinguished Investigator in 2011. Prior to Li and Lim, Wang was named as a GRA Eminent Scholar in 2003.

"These studies are collaborative, multi-disciplinary efforts," Li said. "The support from the GRA has allowed us to build state of the art facilities at Georgia State which have been essential to making this essential life-changing research possible."

Last month's hearings on the constitutionality of health care reform didn't help its popularity: Public support for Barack Obama's signature domestic legislation has hit a new low in the latest ABC News/Washington Post poll, with criticism of the individual mandate as high as ever.

Half the public, moreover, thinks the U.S. Supreme Court will rule on the legislation on the basis of the justices' partisan political views rather than the law. Fewer, 40 percent, think impartial legal analysis will carry the day, with the rest unsure.

Fifty-three percent of Americans now oppose the law overall, while just 39 percent support it - the latter the lowest in more than a dozen ABC/Post polls since August 2009. "Strong" critics, at 40 percent, outnumber strong supporters by nearly a 2-1 margin in this poll, produced for ABC by Langer Research Associates.

Two-thirds continue to say the high court should throw out either the entire law (38 percent) or at least the part that requires most individuals to obtain coverage (29 percent) or face a penalty; just a quarter want the court to uphold the law as is. Those numbers, like views on the law overall, are essentially unchanged from a month ago.

There are political differences: Republicans, who are most likely to oppose the law, are less apt to think the justices will rule on the basis of politics; 41 percent say so, still a not-insubstantial number when it comes to a basic assessment of independent jurisprudence. More Democrats and independents, 55 and 52 percent, respectively, suspect the justices will go political.

As to election politics, the potential repercussions are unclear. Obama's approval rating for handling health care has been more negative than positive steadily since he signed the Affordable Care Act (ACA) into law in March 2010. And the intensity of sentiment has been especially negative, with strong critics exceeding strong supporters by an average of 13 percentage points.

But the president looks better on the issue when compared to the presumptive GOP nominee, Mitt Romney. As noted Tuesday, Obama leads Romney by 10 points (48-38 percent) in trust to handle health care policy. And Romney may have trouble challenging Obama on the individual mandate, given its similarity to provisions in the Massachusetts health care law Romney signed as governor in 2006.

Indeed, among people who oppose the ACA, a relatively tepid 58 percent trust Romney over Obama to handle health care policy. Among ACA supporters, by contrast, 81 percent prefer Obama on the issue.

PARTY and SUPPORT - Given that the debate over the law largely revolves around the extent to which government should be involved in health care, partisanship and associated views on government regulation also play a large role in support for the law.

Seventy-nine percent of Republicans oppose the law in general and nearly two-thirds would like the Supreme Court to throw out the law entirely, for instance, while 63 percent of Democrats support it and nearly half want the court to keep it intact.

Those numbers among Democrats, while sharply different from views among Republicans, underscore the law's weak support. Further, more independents oppose it than support it - 56-35 percent - and 73 percent would like it rejected entirely (41 percent) or insofar as the individual mandate (32 percent), much more in line with Republicans than Democrats.

Similarly, Americans who see over-regulation of the free market as a greater problem than "unfairness in the economic system that favors the wealthy" are nearly twice as likely as their counterparts to oppose the law overall, and nearly three times as apt to want the high court to axe it entirely.

GROUPS - Among other groups, apart from Democrats, support for the law peaks at 65 percent among African-Americans, 64 percent among liberals and 54 percent among the most-educated adults, those who've done postgraduate work; it's also 49 percent among people who identify themselves in the top socioeconomic echelon, upper-middle class or better off than that.

Beyond Republicans, meanwhile, opposition to the ACA peaks at 79 percent among conservatives, 69 percent among evangelical white Protestants and 67 percent of those who say they're in the middle class, but struggling to stay there.

METHODOLOGY - This ABC News/Washington Post poll was conducted by telephone April 5-8, 2012, among a random national sample of 1,003 adults, including landline and cell-phone-only respondents. Results have a margin of sampling error of 4 points for the full sample. The Korea Advanced Institute of Science and Technology (KAIST) announced that a research team from the Department of Materials Science and Engineering has developed a technology that enables scientists and engineers to observe processes occurring in liquid media on the smallest possible scale which is less than a nanometer.

Professor Jeong Yong Lee and Researcher Jong Min Yuk, in collaboration with Professor Paul Alivisatos's and Professor Alex Zettl's groups at the University of California, Berkeley, succeeded in making a graphene liquid cell or capsule, confining an ultra-thin liquid film between layers of graphene, for real-time and in situ imagining of nanoscale processes in fluids with atomic-level resolution by a transmission electron microscope (TEM). Their research was published in the April 6, 2012 issue of Science. (http://www.sciencemag.org/content/336/6077/61.abstract)

The graphene liquid cell (GLC) is composed of two sheets of graphene sandwiched to create a sealed chamber where a platinum growth solution is encapsulated in the form of a thin slice. Each graphene layer has a thickness of one carbon atom, the thinnest membrane that has ever been used to fabricate a liquid cell required for TEM.

The research team peered inside the GLC to observe the growth and dynamics of platinum nanocrystals in solution as they coalesced into a larger size, during which the graphene membrane with the encapsulated liquid remained intact. The researchers from KAIST and the UC Berkeley identified important features in the ongoing process of the nanocrystals' coalescence and their expansion through coalescence to form certain shapes by imaging the phenomena with atomic-level resolution.

Professor Lee said,

"It has now become possible for scientists to observe what is happening in liquids on an atomic level under transmission electron microscopes."

Researcher Yuk, one of the first authors of the paper, explained his research work.

"This research will promote other fields of study related to materials in a fluid stage including physical, chemical, and biological phenomena at the atomic level and promises numerous applications in the future. Pending further studies on liquid microscopy, the full application of a graphene-liquid-cell (GLC) TEM to biological samples is yet to be confirmed. Nonetheless, the GLC is the most effective technique developed today to sustain the natural state of fluid samples or species suspended in the liquid for a TEM imaging."

The transmission electron microscope (TEM), first introduced in the 1930s, produces images at a significantly higher resolution than light microscopes, allowing users to examine the smallest level of physical, chemical, and biological phenomena. Observations by TEM with atomic resolution, however, have been limited to solid and/or frozen samples, and thus it has previously been impossible to study the real time fluid dynamics of liquid phases.

TEM imaging is performed in a high vacuum chamber in which a thin slice of the imaged sample is situated, and an electron beam passes through the slice to create an image. In this process, a liquid medium, unlike solid or frozen samples, evaporates, making it difficult to observe under TEM.

Attempts to produce a liquid capsule have thus far been made with electron-transparent membranes of such materials as silicon nitride or silicon oxide; such liquid capsules are relatively thick (tens to one hundred nanometers), however, resulting in poor electron transmittance with a reduced resolution of only a few nanometers. Silicon nitride is 25 nanometers thick, whereas graphene is only 0.34 nanometers.

Graphene, most commonly found in bulk graphite, is the thinnest material made out of carbon atoms. It has unique properties such as mechanical tensile strength, high flexibility, impermeability to small molecules, and high electrical conductivity. I have been trading currencies for years and have learned a thing or two. It is very addictive actually, and challenging. You certainly do not want to risk your nest egg on retail forex trading. So now you control 100,000 YEN. If the YEN v.s. USD goes up you make money. If it goes down you lose. You can also trade short - betting your key currency goes down. How do you know when to trade? Well, that's the biggest challenge. There are two main methods to trade:

Manually trading
Traders would be actively studying market conditions and charts. Some traders like to trade from charts exclusively - they are called "technical traders". They would detect patterns in the movement of currencies and when they see a familiar pattern that would indicate a reasonable prospect of making a profit - they would initiate a trade. It takes some experience and understanding of what the charts are telling you. I have done that - with mediocre results. Besides, you have to be totally focused. AND to catch the good trading hours. If you are on the west coast it means early morning. Like 04:00. But man is it fun and intense. As soon as you enter a trade your pulse goes up and - well - it's exiting. Not so fun when your currency drops like a rock - but the thrill is there. It does have a gambling feel - which is the biggest reason people lose. They have the gambling mentality. "I'll win it back". Day trading is, or should be a "non emotional" process.

Automated Trading
This is what I now do pretty well exclusively. And it's a whole new can of worms. Most trading platforms these days have the ability to be programmed to execute trades based on some formula. For example, if the moving average moves up by xx steps - buy EUR and so on. Hundreds of these autotraders are out there, and some work - most do not. Problem is - things change. One autotrader system might work this month, but not the next.

However, with a little research and experimenting it is possible to keep your portfolio live and growing.

The best autotraders in my opinion are the ones offered by the trading companies themselves. NOT the fly-by-night operators promising richness. I think if you can make an average of 5-8% of your capital per month you are doing great - and allowing for some losses in the future. Most companies have systems where you can enter in risk tolerance and other variables to pick the best system to autotrade. This is what I did and I think I tried 15-18 systems before I found the right system for my capital and ambitions.

On my account - after some testing and losses - for the past 4 months my little account has slowly grown 14% in total. Which can drop like a stone anytime, but maybe not. To leverage the slow growth (thus proving the system for the time being) I now increase the amount of currencies I trade. So we'll see.

(AP) ? Reigning U.S. Open champion Samantha Stosur put the latest matchup in the Williams sisters' sibling rivalry on hold by rallying past Venus Williams at the Family Circle Cup.

The second-seeded Stosur won five of the last six games for a 6-3, 4-6, 6-3 victory in the quarterfinals on a chilly, windy Friday at the season's first clay-court event. Stosur now has a chance at knocking out both Williams sisters on consecutive days when she meets Serena in Saturday's semifinals.

While Stosur had never beaten Venus Williams before, she's had success against Serena. Stosur beat her to win the U.S. Open title last September and won their only match on clay in the 2010 French Open quarters.

"For sure, clay's not my favorite surface," said Stosur, the 2010 Family Circle champion. "It's always tough playing Serena no matter where."

Serena Williams advanced when Sabine Lisicki twisted her left ankle early in the opening set and had to withdraw after five games.

The buzz for a Williams' sisters showdown had grown all week as the two Americans moved through the draw. Both came into Friday without losing a set and appeared ready to settle some family business at the Family Circle Tennis Center.

This was Venus Williams' first clay-court tournament since 2010 and her second event since a bout with the autoimmune disease Sjogren's Syndrome kept her out of the game since last summer's U.S. Open. Williams won four times last week at the Sony Ericsson Open and had carried that success into the Family Circle quarters.

She could not get past Stosur, though, and won't play again until next month in Madrid.

"I have come so far, so far from the U.S. Open," Venus Williams said.

Stosur shook off some tired legs from an earlier match ? the Australian was forced to come back Friday morning and finish her rain-delayed match from the night before ? and some wayward serves midway through while moving past Williams.

Stosur is seeking her second Family Circle Cup title in three years.

Stosur came out fast for a 5-2 lead in the opening set. Williams mishit Stosur's 113 mph serve on set point.

That's when Stosur's own service game began to fall apart. She struggled to get the ball in play and Williams took full advantage of Stosur's slower second serves. Stosur had the chance to go up 3-1 in the set, but sent a forehand wide. Venus Williams eventually won to tie the set at 2-all and closed it out by winning four of the next six games.

Williams carried the momentum ? along with the support of the crowd ? into the final set and held serve to lead 2-1. Stosur, though, regained her accuracy and Williams began to tire. Stosur only lost two points over the final two games. Willilams slowly walked to the net after her service return went wide on match point.

"I did my best," Venus said. "I know I can play even better. My errors didn't help my cause. At this point, I'm just learning and learning and learning more."

Williams is schedule to play in Madrid, Rome and the French Open as she strives to make the U.S. Olympic team for the London Games this summer. She said her plan was to head home, practice and get stronger for the competition ahead.

She has risen about 60 spots in the rankings from 134th before Key Biscayne to now. "I've never been so happy to be 70th" in the world, Williams said.

Early on, it looked as if Serena Williams might have a fight on her hands as Lisicki stood toe-to-toe with the 13-time Grand Slam winner. However, as Lisicki moved to her left after a shot, she apparently hurt her left ankle and fell to the court. She immediately got up and called for the trainer, who re-taped her ankle so the 22-year-old German could continue.

It was obvious Lisicki couldn't move as she had. She played three more games before retiring in tears. Serena Williams came over to console Lisicki, hugging her near the net and applauding along with the crowd as she left stadium court.

Lisicki was having an MRI. "We'll see what the doctors say and we'll go from there," she said.

Williams has had her share of injuries that forced her to miss major tournaments. She told the young German that she had time to heal and would be back.

"I said, 'It's OK. It's not the French Open. You've got time to get better,'" Williams said.

Ninth-seeded Lucie Safarova ousted No. 4 seed Vera Zvonareva, 6-3, 6-3. Safarova will meet No. 14 seed Polona Hercog in the semifinals. Hercog defeated 13th-seeded Nadia Petrova 6-1, 6-2.
